PrediScreen™ P-gp Toolbox to investigate interactions of small molecules with P-gp (MDR1, ABCB1)

SOLVO applies ATPase and Calcein assays for the identification of molecules interacting with ABCB1 on a large set of early leads. These two assays are optimized for high throughput and detect both substrates and inhibitors of ABCB1 with high sensitivity.

In the case of positive results in either series of experiments, SOLVO applies Caco-2 monolayers as the ultimate proof for P-gp interactions on a smaller set of test articles.

For late development ABCB1 interaction studies based solely on Caco-2 monolayer are also available as a separate package in compliance with the FDA draft guidance.

MRP3 PREDIVEZ™ Vesicular Transport Kit

MRP3 is an efflux transporter expressed on the basolateral membrane of polarized cells. It has an important role in the hepatocytes and in the intestine in the transport of drug and nutrient metabolites to the blood. In hepatocytes the main role of MRP3 is to efflux active metabolites back to the blood and rescue hepatocytes in case of transporter blockade (MRP2 and BSEP) on the canalicular membrane. Inhibition of MRP3 function may lead to liver toxicity. Typical substrates of MRP3 are Phase II drug metabolites with a preference for glucuronides.




MRP5 PREDIVEZ™ Vesicular Transport Kit

MRP5 is an efflux transporter expressed mainly in smooth muscle cells of the human genitor-urinary tract and in blood vessels, including epithelial cells in the heart.
MRP5 transports cyclic nucleuoside monophosphates, such as cAMP and cGMP, and structurally similar molecules, like antiviral and anticancer drugs. The transport of CDCF by MRP5 was described by Pratt et al (Pratt S. Eur J Pharm Sci. 2006 Apr; 27(5):524-32) where CDCF was described as a fluorescent surrogate for Methotrexate, Pemetrexed and 5-FdUMP.



The PREDIVEZ™ Vesicular Transport kit is a simple and powerful tool to investigate drug-transporter interactions. It provides information on any interaction between the ABC transporter and the test drug that would affect the transport of the fluorescent reporter substrate.


PREDEASY™ ATPase Assay Kit for mouse Bsep-HAM

The bile salt export pump (Bsep) is a member of the ATP-binding cassette (ABC) superfamily of transporters mediating canalicular bile acid secretion. Bsep-mediated bile salt transport is strictly ATP dependent therefore stimulation of ATPase activity by a test article indicates substrate nature of the compound. Moreover, drugs reported to be substrates of mouse Bsep as well as cholestatic drugs known to competitively inhibit the transport function of mouse Bsep might be detected in inhibitory type ATPase experiments



PREDEASY™ ATPase Assay Kit for mouse Bsep-HAM is a simple and robust method to investigate interaction between the drug and mouse Bsep transporter. This assay is composed of two tests on the same plate; activation and inhibition study using Taurochenodeoxycholate as a known activator.


Ko134 selective inhibitor for BCRP/ABCG2 membrane transporter inhibition studies

Ko134, the tetracyclic fumitremorgin C (FTC) analog, is a highly potent and specific inhibitor of the ABCG2 (BCRP) multidrug transporter protein. It has a far higher specificity than GF120918, as Ko134 inhibits P-gp with three orders of magnitude less affinity. Its inhibitory potency of BCRP is 25 to 30 fold superior to P-gp inhibition. Due to its non-toxic property at its effective concentrations both in vivo and in vitro, Ko134 is an excellent laboratory reagent.
