Mouse Bsep Vesicular Transport Assay
Taurocholate, the reporter substrate used by SOLVO's mouse Bsep VT assay, is the major endogenous substrate of Bsep. In SOLVO's indirect mouse Bsep vesicular transport assay the interaction of compounds with mouse Bsep is detected as the inhibition of the transport of 3H-Taurocholate, therefore the assay shows the effect of the test drug on the physiological function of the transporter. The assay utilizes Sf9 cell membranes containing mouse Bsep. Expression of the protein is achieved by baculoviral transfection.
MRP5 Vesicular Transport Assay
The endogenous substrates of MRP5 are cyclic nucleoside monophosphates, such as cAMP and cGMP. Therefore, MRP5 may play a role in the regulation of signal transduction by transporting these second messengers out of the cells. As MRP5 is present in blood vessels, inhibition of cGMP transport can elongate the duration of vasodilatation, making MRP5 a target in conditions like hypertonia, angina pectoris, congestive heart failure, and acute pulmonary edema and erectile dysfunction.
SOLVO's protocol for measuring vesicular transport on mammalian cell membranes expressing MRP5 utilizes 3H-cGMP as substrate.
The mouse Bsep and MRP5 Vesicular Transport Assays are either available as a products (SB-mouseBsep-Sf9-VT and SB-MRP5-HEL293-VT membrane preparation) for in house screening or as the Fee-For-Service Screening performed in the SOLVO Screening Laboratory.
