SOLVO’s patented Calcein Assay® and the Hoechst assay are indirect inhibitory cellular assays which provide information on any interaction between the test compound and an ABC transporter by measuring the modulation of the efflux of a fluorescent dye from cells.In the Calcein Assay® the efflux of calcein AM from either MDR1 (P-gp) or MRP1 expressing cells is studied, whereas in the Hoechst assay the efflux of Hoechst 33342 from BCRP expressing cells is studied. Interaction of the test compound with the transporter will inhibit the efflux of the dye and lead to increased levels of fluorescence. Due to their low costs and quick turnover, the dye efflux assays are high throughput, cellular based assays.
